Composition: Ingredients Gms / Litre Proteose peptone 10.000 Sodium chloride 5.000 Disodium phosphate, anhydrous 3.500 Monopotassium phosphate 1.500 Final pH ( at 25°C) 7.2±0.2 Directions: LSuspend 20.00 grams in 1000 ml distilled water. Heat if necessary to dissolve the medium completely.Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. If desired, aseptically add rehydrated contents of 1 vial of EC O157 : H7 Selective Supplement (FD247) for isolation of Escherichia coli O157 from foods to previously molten and cooled to 45-50°C medium . Mix well and dispense ino sterile tubes or flasks as desired pH 7.00-7.40 Store between 10-30°C in a tightly closed container and the prepared medium at 15-25°C.
